Brief Description: The laser scanning microscope 510 META NLO from Zeiss is based on the reflected light microscope Axio Imager.Z1. In addition to an Argon laser and two He-Ne laser, covering the spectrum of visible light for laser excitation, the setup contains a tunable titanium:sapphire laser of the Chameleon Ultra Family from Coherent for multiphoton imaging. The latter emits light from the red and the near infrared in the range of 690 nm to 1040 nm.
Fluorescence detection is accomplished via two photomultiplier tubes (PMT) and the spectral META detector. The latter is an array of 32 PMTs. Fluorescence light is directed onto a wavelength dispersive element and is imaged onto the META detector. Each PMT of the META detector covers a spectral range of 10.7 nm, resulting in a total spectral width of approx. 340 nm (380 - 720 nm). The META detector is a powerful tool for spectral imaging and unmixing. Furthermore, a transmission detector (PMT) detects the scanning laser and provides a transmission light image of the sample.
This microscope can be used for standard 2d and 3d laser scanning imaging, spectral imaging and unmixing and multiphoton excitation. The latter becomes very important for imaging in deep tissues (> 200 µm) or in life cell applications when out-of-focus bleaching is critical. To control environmental conditions during life cell imaging, a perfusion chamber containing the sample can be incorporated into a homemade thermal insulation box with humidity control.
